561nm is usually used alongside or in place 488. It is used in fluorescence excitation for dyes and other similar applications. 488 commonly excites many of the dyes used in cytometry, but a few dyes are pumped more efficiently by 561 IIRC. (375 is used sometimes too I might add) It's also used in confocal microscopy. It basically replaces the Krypton 568nm line in a lot of applications, so anything a krypton ion or a green laser is used for you may see it replacing or working with. It's a really interesting color. A very bright yellow green.
